一字金輪曼荼羅為密教的重要秘法之一，為祈願息災、增益時修一字金輪法之主尊。一字金輪，又稱一字頂輪王、金輪佛頂，以梵語勃嚕唵（bhrum）一字作為真言，為密教大日如來入最勝三摩地所說之真言，譬喻轉輪聖王的金輪最為殊勝。一九一○年指定為日本重要文化財。
畫面中央日輪內的一字金輪佛頂，即大日金輪，手作智拳印，結跏趺坐於蓮座上，蓮座下由七隻獅子承托，頭上繪有華蓋。大日金輪之周圍為轉輪聖王的七寶及佛眼佛母所環繞，下方正中蓮座上承載飾有火焰紋的輪寶，按順時鐘方向，依序配置有珠寶、女寶、馬寶、象寶、居士寶、主兵臣寶及佛眼佛母。每尊像容完整，身形纖細，繪製於鎌倉時代（1185～1333）前期。
